Determining the Hydrophobicity Index of Protected Amino Acids and Common Protecting Groups

Peptides are in great demand the pharmaceutical arena and a majority of these peptides contain 20 or more amino acids. They infrequently synthesised using fragment condensation approach. A key limitation adopting this approach commonly is that protected peptide fragments with high purity often required prior to final steps. It hypothesized understanding hydrophobic nature acids will assist designing optimal purification processes when needed. Whilst myriad hydrophobicity indices reported literature for unprotected acids, lacks any data regarding which form precursor task. In current study, common α-amino sidechain protecting groups were experimentally determined. Different positions each acid within chain considered, namely at C-terminal N-terminal as well internal positions. These give deep insights on respect its position chain. The acquired research facilitated prediction retention time an uncertainty less than ±1.5%.
